That’s why we’ve engineered pioneering second-generation infraluciferase (iLuc) enzymes — engineered to deliver higher activity with infraluciferin and therefore more sensitive clearer and deeper near-infrared (NIR) bioluminescence imaging (BLI) as well as dual-parameter BLI.
Bioflares provide enzyme reporters in third-generation safety lentivirus backbones*. The enzymes are named after minerals that match the colour of their peak emission, with those peaking above 700 nm represented by black minerals.
For dual BLI, we provide Rhodonite and Obsidian luciferases, which have high activities and are spectrally unmixable from each other with both luciferin and infraluciferin. These
vectors co-express different fluorescent proteins via a 2A self-cleavage sequence (for 1:1 co-expression), allowing their discrimination by bioluminescence, fluorescence microscopy and flow cytometry.
For single-colour NIR BLI we provide Jet luciferase, which co-expresses GFP via a downstream internal ribosome entry site (IRES). Both vector types are driven by the human phosphoglycerate kinase (hPGK) promoter and can be used for transient transfection of human cells or for generation of lentivirus by co-transfection with packaging plasmids.
